The controlled release of caffeine from the casein gel tablets has been achieved over release periods lasting from a few minutes to over a few days. A novel casein gel has been acidified at pH of 1.0 as the insoluble controlled-release matrix and spray-dried with coffee for microencapsulation. The optimal inlet temperature of spray drying for the casein-coffee mixtures has been found to be 150 °C. The elastic casein-coffee tablets have been engineered without denaturing the components (as...

Overview graph for the literature and patents on pharmaceutic solid dispersion form 1980 to 2015
20. July 2018
Solid dispersions are an effective formulation technique to improve the solubility, dissolution rate, and bioavailability of water-insoluble drugs for oral delivery. In the last 15 years, increased attention was focused on this technology. There were 23 marketed drugs prepared by solid dispersion techniques. Objective: This study aimed to report the big picture of solid dispersion research from 1980 to 2015. Method: Scientific knowledge mapping tools were used for the qualitative and the...

Process mapping of dry granulation via roll compaction
07. July 2018
In this study, a process map was developed in an effort to improve the understanding of dry granulation of pharmaceutical excipients by roll compaction process, and to implement the quality-by-design (QbD) approach. Through development of the process map, a correlation was made between the critical process parameters (roll pressure, screw speed), and critical quality attributes (density of ribbons and granule size). This method reduces development time, quantity of materials required and cost....

Graphic with overview flow and particulate properties of mannitol and lactose grades
02. May 2018
Appropriate selection of excipient grade during tablet formulation development depends on thorough knowledge in their compaction and flow properties. Each chemically unique pharmaceutical excipientis usually available in several commercial grades that are widely different in powder properties, which influence their performance for a specific formulation application. In this work, 11 grades of mannitol were systematically characterized, in terms of their particulate, flow and tableting...

Overview analysis results on excipients` influcent on the stability of spray-dried powders
23. April 2018
The aim of this study is to investigate the influence of excipients on physical and aerosolization stability of spray dried Ciprofloxacin dry powder inhaler formulations. The model drug, Ciprofloxacin hydrochloride, was co-spray dried with excipients such as disaccharides (sucrose, lactose, trehalose), mannitol and L-leucine. The spray dried samples were stored at two different relative humidity (RH) conditions of: (1) 20% and (2) 55% RH at 20°C. Ciprofloxacin co-spray dried with disaccharides...

In twin screw wet granulation process, the binding excipients could be added in two ways: premixed with powder materials before granulation or dissolved in water as a solution. In this paper, the feasibility of using micronized lactose as a solid binder excipient in twin screw granulation process was examined. Different proportions of micronized lactose were mixed with lactose and microcrystalline cellulose (MCC) powder before granulation, respectively. As a comparison, hydroxypropyl cellulose...

Graphic with process and results to use bosentan microparticles for dry powder inhalation
13. March 2018
In this study, we prepared carrier-based formulations for dry powder inhalers by mixing bosentan microparticles with carrier, prepared in three separate types of lactose. Spray-dried, milled and sieved lactose resulted in formulations with various shapes, surface morphology and particle size distributions.

Development of bi-component generic orally inhaled product (OIP) delivered from dry powder inhaler (DPI) is challenging due to the necessity to demonstrate the similarity of size distribution of both APIs in inhaled aerosol. The effects of selected technical factors on OIP development are investigated. The Monodose inhalers with different aerodynamic resistance were used to aerosolize generic fluticasone/salmeterol formulations. Flow dynamics in the DPIs was analyzed during realistic use. The...

Glucagon is a peptide hormone used for the treatment of hypoglycemia; however, its clinical potential is limited by its insolubility and instability in solution. Herein, the encapsulation, stabilization, and release of glucagon by trehalose glycopolymer nanogels are reported. Methacrylate-functionalized trehalose is copolymerized with pyridyl disulfide ethyl methacrylate using free radical polymerization conditions to form trehalose glycopolymers with thiol-reactive handles.

Propranolol hydrochloride is a nonselective β-adrenergic drug and has been used as angina pectoris, antihypertensive, and that of many other cardiovascular disorders. It has a relatively short plasma half-life and duration of action are considered too short in certain circumstances. Thus, it's fascinating to elongate the action.
