Lipid excipients for suppository and pessary formulation include hard fat and hard fat with additives. These bases provide excellent physico-chemical stability and drug delivery properties covering a wide range of API, including bases optimized for the formulation of Traditional Chinese Medicines. For information on excipient application and formulation processes please see the Formulation Technologies section.

Gelucire® is Gattefossé’s brand of semi-solid excipients originally designed for hard gelatin capsule molding. The name results from the fusion between « gélule » meaning capsule and « cire » meaning wax. Following the name, there is a set of two figures: the first (eg 44) indicates the melting point in Celsius degree, the second (eg 14) indicates the theoretical HLB (Hydrophilic Lipophilic balance). The family is composed of four products with their own characteristics, specificities...

Hot-melt coating is a novel and cost-effective solvent-free coating technique. Commonly used excipients for hot-melt coating are lipid substances. In this work, modified fluidized-bed system was applied for coating, and additional components were used to melt and convey the excipient to the fluidized-bed apparatus. Granules containing 20% (w/w) paracetamol were coated with Precirol® ATO 5 at 9.1% (w/w) coating level. The aims of this study were to define a relationship between the three...

Poly ethylene glycol (PEG) ester surfactants are synthesized by reacting polyethylene glycol with fatty acid. The polyethylene glycol comprises the hydrophilic part of the surfactant and the fatty acid is the lipophilic part. By varying the molecular weight of the PEG and the fatty acid, surfactants covering wide range of hydrophilic lipophilic balance (HLB) values can be produced. Gelucire is the family of vehicle derived from mixtures of mono, di and triglycerides with PEG esters of fatty...

To circumvent the bad taste of drugs, many technologies are available, such as pro-drugs synthesis, formulating with flavors and sweeteners or coating drug particles with a lipid film. The versatility of the latter technology is particularly attractive and at Gattefossé we have worked on developing a simple, efficient and robust taste masking method. Simple We use a standard high shear mixer, very common equipment in the pharmaceutical industry, with a limited number of process parameters to...

The oral bioavailability of poorly water-soluble active pharmaceutical ingredients (APIs) can be improved by the preparation of amorphous solid dispersions (ASDs) where the API is dissolved in polymeric excipients. Desired properties of such ASDs like storage stability, dissolution behavior, and processability can be optimized by additional excipients. In this work, the influence of so-called low-molecular-weight excipients (LMWEs) on the phase behavior of ASDs was investigated.

Lipid-based formulations (LBFs) can be effective oral drug delivery systems for poorly water-soluble chemical entities, provided they are designed with careful selection of the excipients, based on their role in the delivery system and in relation to the drug properties [1]. LBFs comprise four types of formulations (type I, II, III A and B, IV) containing mixture of oils, surfactants and hydrophilic cosolvents, some of which are able to self-emulsify in contact with the gastrointestinal medium...
